During an interview with The A.V. Club, Vik Sahay said that he first auditioned for the role of Morgan, and when the auditions came down to just himself and Joshua Gomez, and Gomez got it, the producers instead offered Sahay the then-smaller role of Lester. Lester has since become a more prominent character than he was in the pilot.


Chuck's alter ego "Charles Charmichael" may be a reference to musician Hoagy Carmichael. Ian Fleming often described his character James Bond as looking like Hoagy Carmichael.


The character of Chuck usually wears Converse Chuck Taylors, often simply referred to as "Chucks".


Natalie Martinez was to join the cast after the pilot episode as Kayla Hart, a love interest for Chuck. Martinez appeared in the cast promotional pictures but her character was written out before she could appear on the show.


A good many references are made to the movie Spies Like Us (1985). The bug used in several episodes is called a GLG-20, which is the designation given to undercover operatives in the movie. Emmett Millbarge is named after the two main characters in the movie, first name taken from Chevy Chase's Emmett Fitzhume, and last name taken from Dan Aykroyd's Austin Millbarge; this is also the order they are introduced in the film. In an episode, Chuck poses as a doctor and references a scene from the movie where doctors go through greeting each other by saying "Doctor, Doctor". Chevy Chase has a guest role in the show.

Daniel Shaw is played by actor Brandon Routh, who has starred as Superman in Superman Returns (2006). At one point Chuck refers to Shaw's physique as "Superman-y".


In the episode Chuck: Chuck Versus Santa Claus (2008) there are several references to the movie Die Hard (1988). The LAPD officer in charge is Al Powell, (and played by the same actor, Reginald VelJohnson), who also eats Twinkies. He responds to the FBI agent who asks "Who's in charge here?" just like in the movie. Al Powell also calls his brother, the store manager, Mike Powell, and uses a line from the film "How Ya holding up in there, partner?"


In Chuck: Chuck Versus the Tic Tac (2010), the box where an important item was kept is number 092407, which is in reference to the date the show premiered, September 24, 2007.


In several episodes Chuck makes or refers to chicken pepperoni which is a dish Chevy Chase made in Seems Like Old Times (1980).


In Chuck: Chuck Versus the Helicopter (2007) when Chuck flashes, he says "Oceanic Flight 815 was shot down". Oceanic Flight 815 was the flight that crashed on Lost (2004).


In the Season 4 premiere episode(2010), Chuck and Morgan constantly use "Tangiers" as their code word to run. This is possibly a reference to The Bourne Ultimatum when Jason Bourne is in Tangiers, Morroco, there is alot of running.


Jeff's full name is Thomas Jefferson Barnes.

When Dolph Lundgren appears Chuck: Chuck Versus the Anniversary (2010) he makes several references to his character Ivan Drago in Rocky IV (1985). He does so when interrogating John Casey telling him, "I must break you," as well as threatening the team saying, "If they die, they die."
